Human brain astrocytoma. 1321N1 is a human astrocytoma cell line isolated in 1972 as a sub clone of the cell line 1181N1 which in turn was isolated from the parent line U-118 MG (one of a number of cell lines derived from malignant gliomas by J Ponten.).

    1321N1 is a human astrocytoma cell line originally derived as a subclone of 1181N1. It was established from a 47-year-old Caucasian male and displays polygonal morphology with adherent, monolayer growth, showing glial characteristics. These cells are low-tumorigenic in nude mice but widely used in neuropharmacological studies. 1321N1 expresses M2, M3, and M5 muscarinic receptors involved in phosphoinositide signaling, forming G-protein-coupled complexes influencing smooth muscle and CNS function. Upon stimulation, they release IP3, trigger intracellular calcium waves, and induce ATP release, facilitating intercellular communication. The line also expresses classic astrocytic markers such as GFAP, vimentin, and S-100 protein. Genetically, 1321N1 is hypodiploid with chromosomal abnormalities and harbors a homozygous TP53 mutation (p.Arg213Gln) and PTEN splice site mutation (c.1026+1G>T). Additionally, mitochondrial DNA mutations disrupt respiratory complexes I and IV, increasing oxidative stress.
