  • Description
    ARPE-19 is a spontaneously immortalized human retinal pigment epithelial (RPE) cell line established from the normal eye of a 19-year-old male donor. These adherent cells retain key morphological and functional features of native RPE, including polarization, tight junction formation, and epithelial monolayer organization when cultured on laminin-coated membranes under low-serum conditions. ARPE-19 expresses hallmark RPE markers such as CRALBP and RPE-65, indicative of its physiological relevance. The cells exhibit a doubling time of 55–65 hours and support up to 48 population doublings, maintaining viability and consistent gene expression across passages. Compared to iPSC-derived RPE cells, ARPE-19 shows elevated expression of genes related to chromatin regulation, eye development, and muscle contraction networks.

    ARPE-19 cells are a widely recognized in vitro model in ophthalmic research, particularly for studying retinal pigment epithelium physiology, ocular drug transport, and retinal disease mechanisms. They are instrumental in modeling age-related macular degeneration (AMD), oxidative stress responses, and RPE-mediated phagocytosis of photoreceptor outer segments. Additionally, ARPE-19 supports transfection and gene expression studies, enabling exploration of pigmentation, retinal metabolism, and barrier function. However, ARPE-19 contains a mixture of mortal and immortal cell populations, and its gene expression profile differs from native RPE cells, therefore, careful experimental validation is advised when modeling retinal pathophysiology or evaluating therapeutic interventions.
