HCEC-B4G12 cell line is a clonal subpopulation started in 2005 from the parental cell line HCEC-12 established from normal cells of the posterior epithelium of the cornea of a 91-year-old Caucasian woman transformed with the early region of the SV40 genome.

    HCEC-B4G12 is an SV40-transformed human corneal endothelial cell line derived from the parental HCEC-12, which was originally established in 2005 from the posterior corneal of a 91-year-old Caucasian female donor. HCEC-B4G12 exhibits epithelial-like morphology with polygonal cell shape and adherent growth characteristics. Genomic analysis confirms stable integration of the SV40 large T antigen, which likely confers immortalization through p53/Rb pathway disruption. Despite this transformation, HCEC-B4G12 retains corneal endothelial differentiation markers. The cells express high levels of collagen type IV and minimal levels of collagen type III, making them a valuable model for studying corneal endothelial characteristics.
