HL-60 is a promyelocytic cell line derived by S.J. Collins, et al. Peripheral blood leukocytes were obtained by leukopheresis from a 36-year-old Caucasian female with acute promyelocytic leukemia.

    HL-60 is a human promyeloblast cell line originally isolated from the peripheral blood of a 36-year-old Caucasian female diagnosed with acute promyelocytic leukemia (APL). HL-60 exhibits lymphoblast-like morphology and grow in suspension. HL-60 has a near-triploid karyotype, typically ranging from 64 to 78 chromosomes. This cell line expresses key myeloid differentiation markers such as CD33 and CD11b but lacks the PML-RARA fusion protein that is commonly present in classical APL cell. HL-60 is tumorigenic in SCID mice, where it can form granulocytic sarcomas following subcutaneous injection. HL-60 possess the capacity to differentiate into granulocyte- or monocyte-like lineages in response to chemical inducers such as retinoic acid (ATRA) or dimethyl sulfoxide (DMSO).
