Description

Human Celiac PBMCs are isolated from the patient’s peripheral blood. Their main components are lymphocytes (including T cells, B cells and NK cells) and monocytes. Morphologically, PBMCs are round or oval. In celiac disease, PBMCs (especially HLA-DQ2/DQ8 restricted CD4+ T cells) trigger abnormal immune responses by recognizing gliadin peptides, releasing proinflammatory factors such as IFN-γ and IL-21, leading to small intestinal mucosal damage. In addition, defective regulatory T cells (Treg) function and activation of cytotoxic T cells further aggravate autoimmune reactions.

Application

Human Celiac PBMCs can be used to analyze T cell activation, proliferation and cytokine secretion (such as IFN-γ, IL-17) induced by gliadin, revealing the Th1/Th17 immune response characteristics of celiac disease. In addition, the molecular basis of antigen-specific immune response can be elucidated by detecting the clonal expansion of HLA-DQ2/DQ8-restricted T cell receptors (TCRs).
