    Human Crohn’s Disease Peripheral Blood Mononuclear Cells are derived from the peripheral blood of patients with Crohn’s Disease (CD). PBMCs, including lymphocytes (T cells, B cells, NK cells) and monocytes.Lymphocytes (T cells, B cells, NK cells) and monocytes are enriched from whole blood by density gradient centrifugation (such as Ficoll separation medium). Abnormally activated immune cell subsets may exist in PBMCs of patients with Crohn’s disease, such as changes in the proportion of CD4+ memory T cells or upregulated expression of monocyte chemokine receptors. Some subsets may exhibit upregulation of chemokine receptors (e.g., CCR2 on monocytes), activation markers (e.g., CD69, HLA-DR on T cells), or transcription factors (e.g., RORγt in Th17 cells), assessed by flow cytometry or qPCR.These cells are involved in abnormal immune responses, including overactivation of Th1/Th17 cells, defective regulatory T cell (Treg) function, and abnormal differentiation of monocytes into macrophages, leading to intestinal mucosal barrier destruction and chronic inflammation.
