    Human Eosinophilic Esophagitis Peripheral Blood Mononuclear Cells (EoE PBMCs) are immune cells isolated from peripheral blood of patients clinically diagnosed with active eosinophilic esophagitis (EoE), typically defined by ≥ 15 eosinophils per high-power field in esophageal biopsies. These cells show elevated activation of CD4⁺ T lymphocytes and myeloid subsets, marked by increased surface expression of CD38 and Programmed Cell Death Protein 1 (PD‑1) compared to patients in remission, gastroesophageal reflux disease (GERD), and healthy controls. EoE PBMCs interact with primary esophageal epithelial cells in co-culture models to upregulate checkpoint molecules such as PD‑1, indicating direct cell–cell immune-epithelial crosstalk. They serve as a valuable in vitro system to investigate T helper 2 (Th2)-driven inflammation, T-cell exhaustion, and novel immunotherapeutic targets.

    EoE PBMCs are used for immunophenotyping and activation profiling via flow cytometry, particularly to assess CD38 and PD‑1 expression on CD4⁺ T cells and myeloid cells in active versus inactive disease states. They enable co-culture assays with esophageal epithelial cells to explore epithelial–immune cell crosstalk and PD‑1 induction mechanisms. Additionally, these PBMCs support humanized mouse modeling for in vivo validation and preclinical testing of immunomodulatory therapies targeting Th2-driven inflammation and checkpoint molecules.
