Cell Name: KO52

Acute myeloblastic leukemia/M2, positive for CD12, CD33, CD2, CD5, CD7, HLA-DR. MDR1 expression.

  • Description
KO52 (K052) is a suspension cell line derived from the bone marrow of a middle-aged male with CD7⁺ multilineage acute leukemia. The round cells express myeloid (CD13, CD14, CD33), lymphoid (CD2, CD5, CD7), and HLA-DR markers. Cytogenetics reveal 17p- with TP53 (codon 248, C>T) and NRAS (codon 13, G>C) mutations. KO52 cells lack BCR-ABL and FLT3 mutations. They can differentiate into monocytes or granulocytes upon treatment with PMA, IL-2, G-CSF, or 1,25(OH)₂D₃, with functional validation. Cultured in RPMI-1640 with 10% FBS, they remain virus-free. In vivo, they form tumors and exhibit extramedullary infiltration. The cells show moderate sensitivity to cytarabine and resistance to imatinib, making them a model for therapy resistance in leukemia research.

  • Application
The KO52 cell line has significant potential in leukemia research. It is a valuable tool for studying the cellular and molecular processes of leukemogenesis and differentiation, revealing the mechanisms of disease progression. Additionally, the K052 cell line can be instrumental in investigating the expression mechanism of the multi-drug resistance 1 (MDR1) gene. By identifying key signaling factors, it offers the possibility of identifying novel therapeutic targets for both acute and chronic myeloid leukemias. KO52 cells can also be used to test antibodies and evaluate the cytotoxic effects of chemicals, aiding the development of new leukemia therapies.
