MCF-12A cells exhibit typical luminal epithelial morphology, three dimensional growth in collagen, and form domes in confluent cultures. The cells are positive for epithelial cytokeratins 8, 14 and 18, and negative for cytokeratin 19.

    MCF-12A is a non-tumorigenic human mammary epithelial cell line originally isolated from the fibrocystic breast tissue of a 72-year-old Caucasian female. MCF-12A exhibits epithelial-like morphology and adherent growth characteristics in monolayer cultures. Crytogenetic analysis confirms a normal diploid karyotype (46, XX) and the cell line is devoid of common oncogenic mutations typically associated with malignancy. This cell line express epithelial markers such as Cytokeratin 18/19 (CK18/CK19), and E-cadherin, while exhibiting low levels of estrogen receptor (ER) and HER2 as assessed by immunohistochemistry. This profile makes MCF-12A a suitable candidate for studying non-malignant breast cancer related disease.
