    Mouse NK Cells are isolated from peripheral blood or solid tissues (e.g., spleen, liver) of healthy mice. As mature lymphocytes, these cells exhibit a round-to-oval morphology with irregular extensions and suspension culture properties. Primary mouse NK cells demonstrate limited proliferative capacity and are typically maintained for 1-2 passages to preserve functional integrity. Key functional characteristics include cytotoxicity against targets cells through perforin/granzyme release and cytokine production. They express surface markers CD49b, CD122, NKG2D, and NK1.1+, with subset-specific expression of Ly49 receptors and CD27 correlating with maturation stages (CD11b+CD27+ for mature subsets). Repeated freeze-thaw cycles should be avoided.
