Under serum-free conditions, these cells retain dendritic morphology and express keratan sulfate proteoglycans, which are critical for maintaining extracellular matrix (ECM) organization and corneal transparency. When exposed to TGF‑β1 or FGF‑2, they activate the Rho/ROCK pathway, upregulate α‑SMA, and downregulate ECM components like keratocan and lumican, thereby differentiating into myofibroblasts. These phenotypic transitions make them a valuable in vitro model for studying corneal fibrosis, wound healing, and drug screening applications in ocular research.
