Cell Name: V79

V79 Chinese hamster lung fibroblasts show fibroblast morphology, stable karyotype, high mutation sensitivity, ideal for genotoxicity and radiation studies.

  • Description
V79 is a Chinese hamster lung fibroblast cell line originally isolated from the lung tissue of a male Chinese hamster. This adherent cell line exhibits a fibroblast-like morphology under standard culture conditions. Karyotypic analysis reveals an aneuploid chromosome count ranging from 20–25, with a stable modal number of 22, making it a preferred model for mutagenicity and genotoxicity studies. V79 does not produce immunoglobulin but expresses Fas(CD95) antigen and demonstrates sensitivity to TNF and anti-Fas antibodies. Due to its genetic stability and high susceptibility to mutations, particularly at loci like hypoxanthine-guanine phosphoribosyl transferase (HGPRT), V79 is widely used in toxicology and radiation research.

  • Application
V79 cells hold significant prominence in DNA repair and damage research, primarily due to their distinct attributes, including a truncated G1 phase, karyotype stability, and the absence of endogenous cytochrome P450 expression. These characteristics make them ideal for gene toxicity assays, especially when used to express exogenous P450 enzymes from diverse species. Furthermore, their suitability for the establishment of stable mutant cell lines proves invaluable in investigating DNA repair enzymes and pathways. Despite certain concerns, V79 cells have played a pivotal role in the study of DNA damage induced by x-rays, UV light, oxidizing agents, and various environmental mutagens, thereby contributing significantly to the understanding of carcinogenic mechanisms.
